30% (V/V) Glycerol, 40 mM Tris-acetate pH 8.3 at 25 °C, 1 mM EDTA, 0.6 % (m/V) SDS, 0.25% (m/V) Orange G

Internal tests revealed that high EDTA concentrations, necessary for stopping restriction digests are correlated with poorly focused bands.

The Simplebiotech 6x Orange DNA loading buffer contains only 1mM EDTA for improved buffer stability during storage. Our 6x Orange DNA Loading buffer contains 0.6 % SDS instead, which efficiently stops most enzymatic reactions.

SDS eliminates DNA interaction with restriction enzymes and DNA binding proteins providing clear and sharp bands. It also suppresses the appearance of additional bands caused by cohesive end annealing.
